Necessary components of an instrument:
The necessary components of an instrument used to measure fluorescence of the sample are:
- Excitation light sources
- Filters or monochromators
- Sample holder
- Detector
- Readout device
Further to the optical elements, most fluorimeters and spectrofluorometers have dedicated computers. These are needed to control the instrumental operating parameters such as, excitation and emission wavelengths, scan rates; monochromator slit widths, detector parameters, etc. These help within the acquisition and post processing of the spectral data.
