What are the limitations of transmission electron microscope?

Limitations of transmission electron microscope:

a. In TEM, the materials need extensive sample preparation; we require producing a sample thin sufficient to be electron transparent.

b. This is a time consuming process

c. The structure of the sample may be changed throughout the preparation process

d. The region of analysis is much small, the possibility which the region analysed may not be characteristic for the complete sample

e. The sample may be damaged through the electron beam, mainly in the case of biological samples.
