Here, a stain has a negatively charged chromophore that is repelled by the negative bacterial surface. It results in indirect staining of microbial cell. It appears as clear area surrounded by coloured background. It is used for visualizing many cell structures like capsule. Negative stains also work better under neutral or alkaline conditions.
