Describes why okazaki fragments are not observed in pcr, Biology

Assignment Help:

Which of the following best describes why Okazaki fragments are not observed in PCR?

A. DNA primase is used for the entire reaction therefore eliminating the production of Okazaki fragments that normally result from the activity of DNA polymerase.

B. DNA ligase is added to the reaction to form the phosphodiester bond among the oligonucleotide primers and the short newly synthesized fragments - on an electrophoresis gel this looks like a long continuous fragment.

C. DNA polymerase has been genetically changed to read the template strands in either the 3' to 5' or the 5' to 3' direction thus allowing for the enzyme to follow the replication fork irrespective of the strand.

D. The DNA template is completely denatured by near boiling temperatures and the oligonucleotide primers anneal to the ends of the fragment that is to be amplified therefore allowing for DNA polymerase to synthesize the new strands in long continuous stretches.
