Assignment:
Intracellular signaling molecule 3′,5′-cyclic adenosine monophosphate (cAMP) plays an important role in signaling cascade. Production of cAMP is controlled through the adenylyl cyclase family of enzymes, which convert adenosine triphosphate (ATP) to cAMP and inorganic pyrophosphate.
As a researcher you goal is to quantitate amount of cAMP present in the test sample that has been provided to you. You supervisor has requested you to come up with TWO independent Approaches (methods) to meet your goal. Your supervisor has provided you with the test sample, pure cAMP and antibody against cAMP. You can make any kind of modifications to the reagents provided (except the test sample) or generate your own reagents.
For each of the two independent methods, please answer the below questions.
Method/Approach 1:
1. Describe the Principle and assay design that you will apply to quantitate cAMP in test sample
2. Types of Controls that generate a High/Max signal, Low/Min signal and Background signal
3. What is expected signal/read out and type of instrument you will use
Method/Approach 2:
1. Describe the Principle and assay design that you will apply to quantitate cAMP in test sample
2. Types of Controls that generate a High/Max signal, Low/Min signal and Background signal
3. What is expected signal/read out and type of instrument you will use.
