Discuss a strategy and provide a diagram if necessary and methodology for overexpression, epitope tagging, isolation/purification, western blotting and confocal microscopy for an unknown alien protein that could be anarobic, and is sensitive to oxygen. It has an open reading frame and encodes a protein that can bind with mitochondrial membranes and interfere with proton generation machinery. Also, design an experiment by which the biological function of the protein can be determined. The biological function should either approve or disprove the hypothesis that this protein inhibits the generation of protons in mitochondria. You will have access to all the instruments, reagents, antibodies and related facilities to conduct the experiment. What is a good method to characterize and determine this proteins mode of action in a biological system either in vivo or in vitro?
