Question (1) You are studying nicotinic acetylcholine receptor (nAChR)-mediated current (InAChR) in a mammalian neuron. You observed that InAchR is decreased by exposure of the neuron to kinaseup, a membrane-permeant drug that nonspecifically activates protein kinases.
You suspect that the decrease of InAchR results from direct phosphorylation of hAChRs by a protein kinase that is activated by kinaseup, although you have not identified the protein kinase involved. You hypothesize that phosphorylation of nAChRs by the protein kinase activated by kinaseup decreases the nAChR single-channel current (the current that flows through the open channel of a single nAChR) and want to test your hypothesis.
