Note: This is a process of elimination. Do not to try to prove your unknown is a specific organism. Your goal is to eliminate as many organisms whose descriptions do not fit your organism as possible.
The Bergey's manual will be the source of information for creating dichotomous keys for the elimination process-do not use other sources without instructor approval.
Part 1: Lab notebook recorded in a bound composition book
All data must be dated & have been recorded at the time performed
The Notebook should contain the following
a. Table of contents
b. Numbered pages
c. Experiments
i. Title
ii. Date
iii. Purpose-what does the test determine
iv. Methods (or reference to page in lab manual)
v. Data - a written descriptions of what was observed in the test: plates, biochemical tests and stains (include magnification), Make sure to include colored drawings (two or more colors) of every result.
These drawings may be supplemented with but not replaced by digital pictures
vi. Results: What do the data tell you about the organism (e.g. is Gram+ or Gram-
vii. Analysis: Groups (to get to a chapter) or specific organisms (genera or species) within a chapter that were eliminated via this test & possible organisms remaining or if the test was ambiguous what invalidated the test.
Part 2: Formal report - Biochemical analysis of unknown
a. Written (not not outline) summary of your test results and why you did particular tests-these should be presented in a manner that explains how you arrived at grouping in the Bergey's Manual, then a genus and then a species as opposed to chronological order.
The this report should identify groups (to get to a chapter) or specific organisms (within a chapter) ruled out by a each test described as well as specific organisms that remain a possibility resulting in a your final identification of your unknown (genus species)
b. Table summarizing unknown biochemical and staining test results (you may use the table from Bergeys here and mark relevant tests for various organisms, including:
A) Cell characteristics
B) Culture characteristics
i. Colonies
ii. Slant
iii. Broth
c. The final page should be a figure (can be hand drawn) of your dichotomous tree for eliminating various organisms.
The report should be typed and not placed in any kind of a binder or cover.
The report summary should be approximately y 2-4 pages of typed information depending on the complexity of your identification procedure
Part Ill: Brief report - Molecular analysis of unknown
Experiment 1
Title : Agar shake
Purpose - to final out my unknown are tolerance
Methods - included agar warmed at 55c with a loop
Experiment 2
Title - gram stain
Purpose - to find out if my unknown is gram positive or negative
Experiment 3
Title - simple stain
Purpose - to find out all arrangement
Experiment 4
Title - endospore stain
Purpose - to find out if my unkown is spore
Experiment 5
Title - catalose
Purpose - to see if catalose is present or not
Experiment 6
Title - Oxidose
Purpose - to see if oxidose is present or not
Experiment 7
Title - Motility deep stab
Purpose - to find if my unknown is motile or not
Streak isolation 1
Purpose - isolate pure culture
Streak isolation 2
Purpose - isolate pure culture
