PART 1: The paper by Shyam et al. will be used to learn about the use of LFIA in detection of Nervous Necrosis Virus (NNV) in populations of fish. The following background information on NNV is provided to give more context for the importance of this application of LFIA.
The virus causing NNV is from the family Nodaviridae and infects many species of marine and freshwater fish. The disease damages nerve cells in the central nervous system, resulting in high mortality. The increasing spread of the virus between wild and farm-raised (aquaculture) fish has stimulated the development of ways to diagnose, treat, and prevent the infectious disease caused by NNV. Of particular importance to the aquaculture industry is the detection of the virus in breeding populations. The virus can be vertically transmitted to offspring so it is important that infected animals be removed from the brood stock.
1. What is your understanding of what point of care testing means in the context of this paper?
2. What advantages does the LFIA technique appear to have over other methods being used to detect NNV in fish?
3. With Respect to Figure 1:
a. Why has one of the antibodies (mAb 2B1) used in the LFIA been conjugated to gold nano-particles?
b. What antigen does mAb 2B1 bind to?
c. Why is monoclonal antibody 2B11 immobilized at the test line in the LFIA strip?
d. Why is anti-mouse IgG immobilized at the control line in the LFIA strip?
4. With respect to Figure 2B:
a. Why did the researchers do the experiment?
b. How did the researchers do the experiment?
c. What are the experimental results (simply describe the results-talk your way through the figure)
d. What do the results tell the researchers about their LFIA test for NNV?
