The enzyme papain is a thiol protease, using a mechanism similar to the mechanism used by chymotrypsin, except that there is an active site Cys in papain carrying out the catalytic role played by the active site Ser of chymotrypsin. There is a His residue in papain that plays the same role as the active site His residue in chymotrypsin.

1) Name the region of the chymotrypsin active site responsible for tight transition state binding in chymotrypsin’s chemical mechanism.

2) Like chymotrypsin, papain should also have a region responsible for binding/stabilizing the transition state in its peptide hydrolysis mechanism. Use your knowledge of the mechanism of chymotrypsin to name the structural element of the transition state that would be tightly bound by papain. (You don’t need to draw a structure.)

What is most likely genotypic arrangement of genes in mother : Red-green colorblindness is the most common form of colorblindness and is caused by two closely linked genes located near the tip of the human X chromosome. Red blindness, or protanopia, and green blindness, or deutanopia, are recessive conditions..
Determine the genotypes of the parents : Black coat color in Cocker Spaniels is governed by a dominant allele and red coat by a recessive allele; solid pattern is governed by the dominant allele of another gene and spotted pattern is recessive.
Use recombinant dna methods to prepare a mutant : A drug company decided to use recombinant DNA methods to prepare a mutant a1-antiproteinase (also sometimes referred to by its old name, a1-antitrypsin) that would be moreresistant to oxidation than the naturally occurring inhibitor, to administer..

Calculate the fraction of the total enzyme active sites : Calculate the fraction of the total enzyme active sites in solution that would be in the active form at pH 7.2. Calculate what kcat would be if the activity of the enzyme were measured at pH 7.2.
responsible for the pH effect on kcat : Suppose that an enzyme uses a catalytic mechanism in which a functional group on the enzyme has to accept a proton from the intermediate formed in the first chemical step
Homozygous recessive yellow-bodied-echinus-white-eyed female : In fruit flies, the yellow-bodied (y), echinus (ec) and white-eyed (w) mutations are all recessive to their corresponding normal alleles. A homozygous recessive yellow-bodied, echinus, white-eyed female (y ec w / y ec w, the gene order is arbitrar..
A woman is heterozygous. : A woman is heterozygous. Which one could produce an egg in which two X chromosomes carry an allele for the recessive trait color blindness: nondisjunction during meiosis 1 or nondisjunction during meiosis 2? Then, Explain.
