Summarize the following procedure: Remove the cell culture media and rinse cells 2x in ~1 mL PBS, pipetting liquid against the wall of the well to avoid dislodging cells Remove the final PBS rinse, and fix cells in 1 mL 2% formaldehyde 10 min at RT*, pipetting liquid against the wall of the well to avoid dislodging cells Remove fixative and permeabilize in 1 mL 0.2% Triton X-100 5 min at RT, pipetting liquid against the wall of the well to avoid dislodging cells Remove Triton and rinse cells 1x in ~1 mL PBS, pipetting liquid against the wall of the well to avoid dislodging cells Remove the final PBS rise and stain DNA with 600 μL 3 µM DAPI, 5 min at RT in the dark**, pipetting liquid against the wall of the well to avoid dislodging cells Rinse 2x with ~1 mL PBS, pipetting liquid against the wall of the well to avoid dislodging cells Remove the final rinse, and add ~1mL PBS for imaging (do not allow cells to dry out), pipetting liquid against the wall of the well to avoid dislodging cells.
