Question: The Meselson-Stahl experiment was repeated for a unicellular eukaryote (2N=6) & we learned that DNA replication is semiconservative in eukaryotes. In that experiment, 15N and 14N were used and monitored in exactly the same way as per Meselson-Stahl (pp37-39, figs 3.1 and 3.2). But, instead of "replication cycles" the scientists had to think in terms of "cell cycles" because they were studying a eukaryote that produced cells as per the cell cycle (pp 50&329, lecture notes, and Biology 212). After growing the cells in only 15N for several cell cycles, the scientists transferred the cells to 14N medium (no 15N in the medium) at the G1 phase to start the experiment; that G1 phase is the start of "cell cycle 1". The cell cycle consists of phases G1, S, G2 and Mitosis; in that order.
