4. A student collected a bunch of fresh leaf Pinus sp. from a hillside. He would like to make microscopic observation of the internal tissues of the leaf and study the plant proteins extracted from the leaf. He is interested in some special proteins found in the protein extraction. The molecular weight of the proteins of interest are in the range of 50-100 KDa.

Students should write a proposal of experimental design including the materials preparation and experimental procedures to achieve the following purposes.

i) To prepare the slides for microscopic observation under a bright field microscope.

ii) To determine the total concentration of proteins extracted from the leaf.

iii) To apply an electrophoretic methodto check whether the proteins of interest (the molecular weight in the range of 50 - 100 KDa) are presence in the protein extract of leaf.
