Prokaryotic Identification and Flow Charts

Many different characteristics are used to identify bacteria. Microbiologists often depend upon flow charts to arrive at a preliminary identification for an organism. Flow charts are created from known results concerning bacteria. When unknown bacteria are subjected to a battery of tests the flow chart can help direct and minimize the investment of time, energy and media.

The flow chart is no good if the appropriate organisms are not included. Another important factor is that you must flow down the chart, you cannot skip to individual tests.

You are given an unknown bacterial culture. The result of the gram-stain for the bacteria showed purple color of coccus species that arranged in clusters (under 1000X, LM). You then perform a test on the bacteria using mannitol salt agar. After 24 hour incubation at 370C, the result showed the red mannitol salt agar plate turned yellow.

A) What is the scientific name of the bacteria?

B) Discuss with your classmates how did you conclude the name of the unknown bacterial culture.
