Enumeration of bacteria
Aim
To prepare serial dilutions and perform a spread plate technique to enumerate the number of 'table bacteria present in a bacteriological sample.
1. Label 8x serological tubes and number these 1-8.2. Pipette 4.5ml of sterile saline into all tubes.3. Add 05.ml. of Ecoil to tube 1 Mix with a vortex. What is the dilution of tube 1? 4. Perform a 1/10 dilution from tubes 1 to 8, by transferring 0.5mL from tube 1 to tube 2 etc., mixing well after each dilution.5. Label the 6 PCA plates with your initials, date and the dilutions (10'3 to 108).6. Add 0 lmL of the appropriate dou'ion in your PCA plates and prepare a lawn culture as demonstrated by your supervisor.7. Incubate the plates at 35°C for 24-48 hours.
Please note: Pipettes should be discarded directly into the Biohazard bag and at no time should they be left on the bench or placed in a beaker.
