Position of the amino acids in the tertiary structure of the protein does not change much when it is kept in the aqueous solution after the mutation. When a protein is kept in aqueous solution, all the hydrophobic amino acids will be hidden inside of the tertiary structure meanwhile, all the hydrophilic amino acids will be exposed out to the aqeous environment. Mutation mentioned in the question states that all the lysine residues are replaced with aspartic acid residues. Both the amino acids are hydrophilic in nature. So, in aqeous environment, they will be exposed out. Therefore, mutation does not change the positions of amino acids.
