You discover that a gene for resistance to a new antibiotic is rapidly spreading through a pathogenic population of Pseudomonas aeruginosa. You find that in culture conditions where the bacteria can't grow because they are missing an essential nutrient, the gene still spreads through the population, indicating that it is spreading horizonally. Unfortunately, your DNA sequencing facilities have gone down for the week, and your Dean expressly forbids you from spending money on outside sequencing resources. Therefore you have to solve this problem "Old School." But you have other modern molecular biology tools, and the ability to make a marked recipient strain for experiments.
