Glucose oxidase is an enzyme that can be used for measurements of glucose levels by combining this reaction with an oxygen probe. By sandwiching the enzyme between two gas permeable membranes, the oxygen diffuses toward the dissolved oxygen probe, and its associated removal rate is proportional to the reaction.
a. Give an enzyme on either side of this from KEGG.
b. Write down a "reasonable" expression for the enzyme kinetics of this enzyme.
c. Describe the role of this enzyme in honey.
d. If a pure enzyme preparation is available, what could limit its use for these applications.
2. Calculate the time it would take to oxidize 90% of 2 g glucose/L using 1 mg/L enzyme in free-solution (not the sandwich above), and conduct this assessment based on information from two sources:
a. the first entry in BRENDA for Km at the conditions used for glucose oxidation that were used to make this kinetic measurement. Note the first entry for glucose is Pleurotus osteatus.
b. Aspergillus niger glucose oxidase available from Sigma-Aldrich.
