Lab - Spectrophotometric Quantitation of β-Carotene

1. Using your data for the standard β-carotene assay, construct a standard curve by plotting A449 (y-axis) vs. mg/ml β-carotene (x-axis). This may be done by hand using proper graph paper (20 squares per inch) or by computer. Tabulate your data on the back of your graph.

2. Give a sample calculation (with units) for the final β-carotene concentration in one of the standard tubes (B-E).

3. Calculate the mg of β-carotene per g carrot (mg β-carotene/g carrot). Indicate your method neatly and clearly, including units. (The following steps will helpful).

a. Calculations for ε from standard curve.

b. Concentration of β-carotene in diluted extract.

c. Concentration of β-carotene in concentrated extract.

d. Total mg β-carotene extracted.

e. mg β-carotene/g carrot.
