You have been given an unknown bacterial culture with at least two different bacterial species contained in it. At your disposal, you have the following supplies:
Stains: Gram stain, Negative Stain
Growth Media: Mannitol Salt Agar, MacConkey Agar, Levine Eosin-methylene Blue Agar (EMB), Tryptic Soy Agar (TSA), Nutrient Agar, Blood Agar Using only these supplies, devise a strategy to
1) Isolate the two different bacteria in pure culture (how and on what media?).
2) Begin identification of the two bacteria (how and on what media; what would results indicate? Hint: if you look at the different media available, what results would you expect from them?).
Hint: How can staining help you determine which media to use to identify the microorganisms? How and when would you apply staining?
