Question

You are using a 1.6 cm internal diameter column as a scale down model to purify a Fc-fusion protein. The Protein A resin used is the Mab SelectSure LX, which can provide a dynamic binding capacity (DBC) of 55 mg of your molecule per mL of resin, at 10% column breakthrough. As part of the downstream process development, your team need to prove the method reproducibility in pilot scale. Then, a 200-L bioreactor operated in fed-batch mode will be available, providing you 160 Liters of cell-free media (CFM) material to be purified. The process analytics department confirmed the titer in the CFM is 2.2 g/L. Considering the DBC above, calculate the minimum resin volume necessary in the packed capture column to perform the purification. ( what other information is needed ?)
