Question:

You have been tasked with identifying the microorganism in a sediment that is responsible for the consumption of a toxic pollutant, dichloroethane (DCE). It is known from work in other environments that the key gene in this process is called an alkane dehalogenase. Fortunately, there is a chemical assay for the production of the halogen gas (chlorine, highly toxic!) that allows you to monitor this process. There are also substrates that can be used in the lab to safely grow bacteria on DCE as a sole source of carbon and energy. Describe enrichment and isolation experiments you would do to obtain this microbe. Describe an experiment using either loss of function or gain of function mutagenesis to identify the gene responsible for this activity?
