Questions: To your annoyance, your P.I. now asks you whether you also ran the protein on a non-reducing SDS-PAGE. You said no and grudgingly decided to run the same protein gel as before but now add one protein sample in which you added reducing agents. Because you were annoyed at the request, however, you forgot to label your tubes. Now you don't know which one is which. Do you repeat the experiment, or do you actually know which one is which and if so, why? Your P.I. sees the result and is quite concerned given that you want to do activity measurements with this enzyme.
