Preparation of Solutions

Instructions: Please send the calculations of the exercises

Answer the following solution preparation exercises. You must place all the calculations and the final results. Remember that the solutions are prepared with water, therefore, you must calculate the amount of water that is required for each one.

1. You have a 1M Tris solution and a 0.5M EDTA solution. How much of each will you need if you are going to prepare 1L of TE buffer (10mM Tris, 1mM EDTA)?

2. Prepare 225mL of a 1.5M sodium dodecyl sulfate (SDS) solution.

3. You are given 2 containers with two different reagents. In one of the containers there are 500g of SDS and in the other there are 325g of NaOH. You are asked to prepare 155mL of a solution containing 20% SDS and 2M NaOH. How many grams will he need of both reagents?

4. Prepare 833mL of a 15% NaCl solution.

5. In an agarose gel electrophoresis experiment you should prepare 30mL of a 1% agarose gel and 250mL of 1X Tris-Borate-EDTA (TBE). You are given a container with 50g of agarose and 1L of a "stock" solution of TBE 10X. Perform the calculations to prepare the gel and electrophoresis buffer.

6. You are to prepare 500mL of a 1 in 49 dilution buffer. The solution is to be diluted with water. How many milliliters (mL) will be needed of the buffer and water to prepare the solution?

7. Calculate the molarity of a 9.5% KOH solution.

8. You are asked to prepare 386mL of nutrient agar culture medium. He is given the container of the medium and in the recipe it indicates that the medium is prepared by pouring 32g into 1L of water. Make the calculations and indicate how many grams you are going to weigh the medium and how much water you are going to measure.
