Problem- You are interested in how tRNAs might be inhibited in vivo to the addition of high concentrations of the metabolic cofactor FMN which normally should not be a problem for a cell. But you notice that relatively high concentrations of FMN, specifically above 100 ?M and at elevated temperatures, appear to inhibit E. coli growth and you have traced it to some sort of interactions between FMN and e coli glutamine tRNA. Thus you decided to do an in vitro SELEX experiment, starting with the wild-type E. coli tRNA Gln( CUG) sequence, to discover strong FMN aptamers at 48 ° C. Describe in detail how you would carry out these experiments and comment on whether the results that you get might have any beating on your observations of growth inhibition in vivo or not.
