It is desired to determine the micrograms of thiamine, vitamin B per gram of pork. A 2.00 g sample of pork was extracted with HCl, treated with phosphatase, and diluted to 100 mL. An aliquot of 15 mL is purified by absorption and elution during which process it is diluted to 25.0 mL. Of this, two 5-mL portions are taken, one of them is treated with Fe(CN)63- and both are made up to 10.0 mL for fluorometric examination. A standard solution of thiamine containing 0.2 micrograms per mL is subjected to similar treatment, except that the portion introduced into the absorption column is made up to its original volume after elution (i.e, not diluted). Two 5 mL aliquots are taken, one is oxidized and both are made up to a final volume of 10.0 mL for measurement by fluorescence.
