Question

To express human protein RNAse H1 in E. coli, a researcher isolated the cDNA coding for the protein and transferred this DNA into an expression vector. The vector (plasmid DNA) contains a promoter followed by recognition sequences for the restriction enzymes Nco 1, BamH 1 and EcoR 1 (aligned in this order) for the cloning of DNA fragment containing the RNAse H1 coding region. Describe a strategy to prepare the DNAs and transfer the cDNA fragment coding for RNAse H1 into the expression vector DNA.
