The proteins in the mixture are separated by chromatography. You have reason to believe that your unknown protein is an enzyme which catalyzes the cleavage of phosphodiester bonds. You have a solution of bis- nitrophenylphosphate (BNPP), a phosphodiester compound, which is cleaved to produce nitrophenylphosphate (NPP) and nitrophenol. NPP absorbs at 400nm. Describe how you would test the fraction from the chromatography experiment to determine if they contain the enzyme.
