1. Explain briefly what the purpose of a negative control is.

2. Imagine a chemical reaction as follows: The enzyme alkaline phosphatase (AP) mixed with the two colorless reactants, 5-bromo-4-chloro-3'-indolyphosphate (BCIP) and nitro blue tetrazolium (NBT), produces a purple-colored product.

If you were measuring the effect of enzyme concentration on product formation you might have five tubes with different dilutions of the enzyme AP. You would then add the reactants and measure the time it takes for a certain color change to occur. What would an appropriate negative control be? There may be more than one.

3.Explain why a competitive inhibitor can be overcome by adding more substrate to a reaction but a non-competitive inhibitor cannot.

4.Do you think that a competitive inhibitor could be overcome by adding more enzyme? Why or why not?
