Question

Tonegawa showed DNA in a myeloma cell, encoding the constant and variable regions of the expressed kappa chain, were joined in the DNA from the myeloma cell but were further apart in DNA from embryo cells. He used v and vc probes and southern blots. Tonegawa had to use strick conditions of probing so that his probes did not anneal with kappa v regions, present if different DNA fragments, sharing considerable homology but not identical with the expressed kappa v gene segment. What would have happened if he used less stringent conditions? Suppose he had done his experiments with lambda light chain expressing myeloma cells, using the appropriate lambda v-c and c probes. How would his findings be different from the previous situation when he used less stringent conditions when using kappa probes? Justify your answer.
