Suppose you are studying a new fungal pathogen Candida ickius that is highly resistant to the azole class of antifungals and you note that there is an efflux pump, Pmp-1, that appears to be at very high levels in the cell membrane; this is unlike the very genetically similar organism Candida noticky which is sensitive to the azoles and has low levels of Pmp-1 in its membrane (as determined through comparative western blotting). You also recall that Dr. I.M. Theman published that pmp-1 is transcriptionally repressed by Not-1. Western analysis of Not-1 in both Candida ickius and Candida noticky indicates that Not-1 is expressed much less in C. ickius.

How may this impact drug resistance in C. ickius versus C. noticky? How would you design experiments to explain the differences in resistance between the two strains to the azoles?
