Assignment:

1) What are the similarities and differences in the mechanisms by which macrolides, such as erythromycin, and rifampin inhibit their bacterial targets? What are the similarities and differences in mechanisms of resistance that develop spontaneously to macrolides and rifampin?

2) Compare and contrast the activity of penicillin and clavulanic acid with penicillin-binding proteins and with serine beta-lactamases. Based on this mechanism, how is clavulanic acid used therapeutically? Would you expect resistance to clavulanic acid to develop in clinical bacterial pathogens? Explain your answer.

3) Compare and contrast the signaling pathways that lead to induction of the β-lactamases BlaZ in Gram (+) Staphylococcus aureus and AmpC in Gram (-) Escherichia coli when bacteria are exposed to β-lactam antibiotics. Emphasize the signal molecules and points of negative repression and positive activation in each mechanism.
