Detailed Question: what I am looking for is the protocol only for using gold nanoparticle and also for phosphate and chitosan just protocol experimental protocol

Study Title: Study Title: Use and measuring ultrasound-mediated in cell to detriment nanopartical transport .

The hypotheses to be tested include:

1) change in ultrasound frequency in cell with nanopartical and control sample without ultrasound.

2) measure of two sample to determine the measurement.

3) The beneficial response of ultrasound in cell using nanopartical

4) The cell reaction in different days to measure the affect if ultrasound

Hypothesis -

The aim of this study is determine the efficacy of using Ultrasound technique in improving nanopartical movement and measure it.

- The specific objectives of this study are:

1) To determine the change response of ultrasound in cell with nanopartical - The percent of cell effected by ultrasound with percent of cell not effected by ultrasound. To detect any increase in cell level.

2) To determine which specific wavelengths can increase the nanopartical movement inside cell.

3) To determine mechanism responses in both affected and non affected cell.

4) To determine the time course of movement in nanopartical.

Research Design and Methodology
- Research Design

• Ultrasound applicator Model:27383:frequency 1.0,3.3 Mhz, intensity 0-2.5

• Cell :MC3T3 or L929

• Nanopartical: gold or silver

• Geliten :REQVSL:34564

- Data Management and Analysis

-the statistical methods to be used for our study is two independent sample t-test.

- Method:

Gelatin from bovine skin obtained fromUniversity lab.
Cylindrical or cell well plate? (smple 1-18)
Nanopartical will filed in 1-18 sample

Experimental:

Ultrasound: the focus of ultrasound beam will position at cell well plate or cylindrical in sample 1-9 the second sample pairs 10-18 will immersed but not expose to ultrasound and will act as control then, sample will be taken for histological purposes to evaluate the different sample.

Detection of ultrasound: sample will determine using microscopy.

Title of Directed Study: Effects of ultrasound on transport of gold nanoparticles through biopolymer hydrogels.

Summary (of the contents and topics covered in the directed study)

• Literature review on uses of gold nanoparticles as contrast, drug delivery, diagnostic, and therapeutic agents, and on the use of ultrasound to enhance nanoparticle transport in thick tissue

• Preparation of gold nanoparticles of sizes 6-100 nm, and hydrogels with varied physical composition.r

• Preparation of a study plan

• Tracking of gold nanoparticle transport within hydrogels by microscopy

• Optimization of ultrasound parameters (time, duty cycle, frequency, energy), used to enhance transport of nanoparticles into tissue.

• Preparation of a scientific report describing and interpreting the experimental methods and results.
