A) If you were given an unknown bacteria which you stained and looked at under the microscope and found gram positive rods, according to our charts, what bacteria could it be? How could you differential them? What is another visible characteristic found in these bacteria that you could see under the microscope?

B) Looking at the tests for the gram positive bacteria we use in lab, which one has a characteristic not found in the others? What is this used for?

C) A few of the gram positive bacteria do not have catalase. Yet they are common in our environment and some account for much disease world-wide. How do you suppose they can still survive in an oxygen environment?

D) Your instructor gives you an agar plate full of loose and flowing colonies, and tells you it is your unknown to identify. You carefully sniff the plate and find the smell is like a dirty diaper. Now you have a big clue as to the identity of the bacteria. What further test might you do to decide the genus and species?

E) Looking on the identification chart in the lab manual, why are the fermentation tests negative for the Kocuria rosea? That is, what kind of metabolism does this microorganism have?

F) Notice that sometimes on the identification chart the cell for describing a bacterial test as positive or negative just has a V for variable. Describe why this happens.

G) The urease test listed in the lab manual is only done as needed because we have to buy instead of make this tube ourselves. What bacteria have you narrowed down you identification to if you have to ask for this test?

H) Why do you suppose there is no column for hemolysis on the gram negative chart?
