In my Biochemistry lab this week we made buffer solutions. I had TRIS. I made 100ml of .1M solutions. The original ph was 9.15. I added HCl until ph matched the listed pka of 8.3. weight of TRIS is 1.235g.
(a) what is the ratio of A- / HA in your buffer are present in the solution?
(b) How many micromoles of A- and HA are present in the solution?
(c) If you now add 3ml of 1M NaOH, will you still have a valid buffer? Explain.
Calculate the theoretical ph of one of your buffer at 0^0 C. Assume that room temperature is 22^0 C. If none of your buffer is listed on the table of changing pka with temperature, do this problem for TRIS at ph 8.0.
