Practical - Agglutination and Complement-mediated Lysis

Learning objective 1: Demonstrate accurate pipetting skills which will be needed to complete a demonstration of antibody mediated agglutination of sheep red blood cells and of complement mediated lysis

Experiment summary
Aim: This experiment examines the ability of polyclonal antibody production to aid in immune defence by coating cells expressing foreign antigen resulting in agglutination (cross-binding) and complement mediated lysis.
In this practical you will:
Prepare a single cell suspension of sheep red blood cells (SRBC) in Alsevier's solution;
Titrate Rabbit-anti-SRBC into the SRBCs & observe the optimum ratio of antibody to RBC for agglutination;
Titrate in plasma from various sources containing active complement factors and observe the optimum ration for RBC lysis.

On one side of an A4 page (no more than 250 words) discuss the mechanisms by which antibody and complement contribute to the elimination of a pathogen from the blood stream. For the purpose of this exercise, you may assume that the antibody is already present. Submit your written report to the demonstrator before leaving the prac.
