You need to make tubes containing the following dilutions of milk: 10-4, 10-5, and 10-6, from which you will plate 0.1 ml aliquots, using the pour plate technique. Make a well-labeled flow chart for how you would perform the feat. You must transfer 1.0 or 0.1 ml aliquots into your dilution blanks and you can only use 9.9 ml or 9.0 ml dilution blanks.

2. Your friend has an E. coli culture diluted out to 10-8. He plates 0.1 ml of the culture on each of three TSA plates. The next day, you count the colonies and there are 180, 141, and 163 colonies on the plates. Calculate the CFU/ml of the original undiluted E. coli culture.
