Instructions
You are required to write a 1500-word report about specific practical sessions/experiments.
The report must include each of the following parts (marking criteria &suggested word count for each section are shown in brackets):
Part A: Enzymatic activityof one natural and two cloned AMPD1 enzymes(350 words)
You will conduct an experiment (Practical 6 - Determination of the Activity of Human AMPD1 Enzyme). Following this session, you will be required to conduct another experiment (Practical 7 Determination of the activity of cloned and human AMPD1 enzymes) and propose a hypothesis about the activity of one natural and two cloned AMPD1 enzymes and design an experiment to test your hypothesis. You will need to perform analyses of AMPD1 enzymes from a variety of sources to determine the Km and Vmax.
• Hypothesis and Aims of the experiment - state the aims that the experiment(s) set out to test and formulate a hypothesis
• Materials and Methods - provide details of how the experiment was carried out and what materials were used. These details should have been recorded in your laboratory notebook.
• Results - present data including a data analysis followingthe steps: observation, interpretation, and conclusion.NB: No statistical analysis is needed for this part.
• Brief conclusion statements - a commentary on the results from the part A section
Part B: Yeast and Bacterial expression systemsto produce AMPD1 enzyme
Similarto the statistics exercise during workshop, you will need to perform appropriate statistical test(s) to determine if there is any significant difference in the production yield of the enzyme from each organism.
You will need to integrate these results into your laboratory report assignment.
Use the information and guidance you have been given in the statistics lectures and workshops (including the model answers) to help you.
You will be provided with expression data in excel to perform an appropriate statistical analysis using SPSS.
Experimental design:
Experiments were carried out to compare the amount of AMPD1 enzyme which bacteria and yeast can produce (the ‘yield'). To do this, we created yeast and bacteria which were able to produce the enzyme. We then set up ten identical cultures of the bacteria and ten identical cultures of the yeast and grew them overnight. The next day we harvested the cells and collected the enzyme. We then calculated the concentration of extracted enzyme from each culture.

• Brief conclusion statements - a commentary on the results from the part B section
Part C: Discussion and Conclusion (400 words)
This section should highlight the main findings of both parts and discuss them in the context of other published studies.
Based your analysis issued from both parts, you shouldconclude the report by discussingwhich enzyme you would selectto carry out the industrial production in the biotech company.
References - in-text citations and bibliography presented in the Harvard style (does not contribute to word-count)which should only be presented in the introduction and discussion sections.
